Henri Sarles est né en 1922 à Ermont (Val-d’Oise). Il a mené ses études secondaires au lycée Périer à Marseille et ses études supérieures aux facultés des sciences et de médecine de Marseille.
Externe, puis interne (1946) des hôpitaux de Marseille, chef de clinique, maître de conférence.

Docteur en médecine (1950), médecin des hôpitaux (1955).
Doctorat d’Etat ès science de la faculté des sciences de Marseille. Thèse dirigée par le professeur Desnuelle (1954) sur les phosphatases alcalines.
Intéressé par la recherche en biochimie, Henri Sarles travaille dans le laboratoire de Jean Roche, professeur au Collège de France (1948-1957), menant de front recherche et activités cliniques.
Chef du service de gastro-entérologie à l’hôpital Sainte Marguerite, à Marseille (1959-1992).
Directeur de l’unité de recherche Inserm 31 « Pathologie digestive » (1962-1984).

Travaux scientifiques
Les travaux d’Henri Sarles, internationalement reconnus, ont porté pour l’essentiel sur le pancréas et les voies biliaires.
Sans moyens, dans une ville sous-développée comme l’était Marseille à la fin des années 1940, Henri Sarles a commencé ses recherches par l’étude statistique simple de cas avérés de pancréatites. Il a démontré, pour la première fois, que les pancréatites aiguës, causes présumées des pancréatites chroniques par leur répétition, survenaient en moyenne 15 ans après celles-ci, ce qui infirmait ce qui était admis jusque là.
Par ailleurs, des études morphométriques et statistiques des lésions aux échelles optique et ultrastructurale lui ont permis de montrer que la première lésion de la pancréatite chronique la plus fréquente, c’est-à-dire la pancréatite chronique calcifiante, était la précipitation dans les canaux pancréatiques, avec de la calcite (CaCO3), d’une glycoprotéine, cette dernière ayant été découverte, purifiée et séquencée dans son laboratoire. Cette glycoprotéine, d’abord appelée PSP puis lithostathine, est diminuée ou, plus rarement, absente dans le suc pancréatique des malades atteints de pancréatites chroniques calcifiantes. La lithostathine joue un rôle prépondérant dans la stabilisation du calcium dans le suc pancréatique, rôle qui a été précisé par Henri Sarles et ses collaborateurs. D’autres facteurs lithogéniques (hyperconcentration des protéines sécrétoires et de la lactoferrine, baisse de concentration du citrate) ont été observés dans le suc pancréatique des patients.
Les causes les plus fréquentes dont les lésions sont identiques sont : 1) l’alcoolisme associé à un régime riche en protides et en lipides, dont la relation statistique avec le risque de la maladie a été calculée ; 2) la carence protéique de la mère en gestation et de l’enfant. Ces facteurs ont été étudiés expérimentalement chez le chien et le rat, et chez l’homme dans 17 pays (Inde, Afrique, Europe).
Le probable rôle des chocs psychiques dans la survenue d’oddites chez l’homme a été montré et la maladie reproduite chez le chien par irritation chronique du nerf splanchnique droit.
Des études physiologiques ont démontré, pour la première fois, l’existence d’un mécanisme inhibiteur de la sécrétion pancréatique déclenché par le contact des glycérides avec l’iléon ou le c‰ìcum selon les espèces.
Les recherches d’Henri Sarles et de ses collaborateurs ont donc toujours été menées à partir de constatations cliniques et/ou physiologiques, passées au crible d’analyses statistiques, puis étudiées par les techniques morphologiques ou biochimiques adaptées.
Henri Sarles a eu plus d’une cinquantaine d’élèves étrangers et six professeurs nord-américains ont passé leurs années sabbatiques dans son laboratoire.
      Les travaux d’Henri Sarles et de ses collaborateurs ont donné lieu à plus de 600 publications scientifiques dans les revues à comités de lecture.
